Synthace’s deferred transfer functionality allows you to delay the addition of any liquids in a specific element. This is particularly useful when you wish to have a device add a specific reagent, but use another device or manual pipetting to carry out the rest of the experiment. Overview
To start, correctly configure the dilute element by connecting diluents and liquids to dilute as element inputs.
To specify a deferred liquid transfer in a dilute element, toggle to Delay Additions and open up the dropdown menu.
If you select None, both diluents and dilutees are added by this element.
If you select Dilutee, then only diluents are added by this element.
If you select Diluent, then only dilutees are added by this element.
If you select Dilutee and Diluent, then this element will do nothing and dilutions should be completed using Mix Onto elements.
In this example, compounds are diluted in media. Compounds are added at extremely low volumes so a Beckman Coulter Echo 650® is used to first dispense the compounds onto the assay plate, and then media is added via manually pipetting. Since the media is the diluent of the dilutions, Diluent is selected under delay additions.
Describe the rest of the dilution parameters such as Final Target Volume, Target Concentration and Replicates.
Since diluent liquid transfers are delayed, Synthace will warn users that an additional downstream element is required to complete the workflow.
Add an additional stage specifying Manual mode and a Mix Onto element in the new stage. Wire the dilutions as the Destination Liquids of the Mix Onto element and the Remaining Diluents as the Source Liquids of the Mix Onto element. Additionally wire the Remaining Transfers from Dilute to the Transfers to Complete input of Mix Onto.
There is no additional information needed to specify in the Mix Onto element since Synthace already knows of all the remaining transfers it needs to complete.
Once complete, simulate the workflow. Synthace will add all compounds (liquids to dilute) in the first stage of the workflow using the Echo® and then will add the Media (diluent) manually in the second stage of the workflow.
