I invited a user to be a part of my worship plan. She didn’t respond to my email, but I know what her response is. Can I mark this myself?

You sure can. Go to your worship plan and click "Show Responses" (assuming you checked "Team Member Confirmation System" when you created the plan) and click "Manually Confirm People."
