Use unselected liquids

Core elements tutorial 8.3

In Core elements tutorial 8.2, we demonstrated how you could select liquids based off of a tag that those liquids have assigned to them and aliquot them to a new plate, in this example all liquids with the tag Sample.

Following on from that tutorial we will show you how you are able to also use any liquids that were not selected at the same time, in this example diluting the standard defined on Plate A.

Get started

  1. Read the introduction to this series of tutorials, then complete all of the previous tutorials in the series. For more information, click here.

  2. Create a copy of the workflow that you created in Core elements tutorial 8.2. To learn how, click here.

  3. Rename the copy Core elements tutorial 8.3. To learn how, click here.

Dilute unselected standards on the assay plate

  1. In the Workflow Builder add another Define Liquids and Plates element and a Dilute element to your previous workflow (already containing 1 x Define Liquids And Plates, 1 x Select and 1 x Aliquot elements).

  2. Connect the Unselected Liquids output parameter from your Select element into the Liquids To Dilute input parameter of your Dilute element.

  3. Connect the Liquids output parameter from your second Define Liquids And Plates element into the Diluents input parameter of your Dilute element.

  4. Define a diluent you want to use for diluting your standards in the second Define Liquids And Plates element. Here we have done this via the Parameters option.

  5. Specify the dilution parameters you want for your standards. Here, diluting to a New Location, by Serial dilution and By Factor with the following parameter values.

Simulate the workflow

  1. Check that the device that you selected can follow the instructions that you prepared. To learn how, click here.

Preview the execution

  1. After you simulate the workflow, click View Simulation to open the simulation details.

  2. Open the Preview tab, then click through the steps to check that the instructions that Synthace has generated are correct.

    Note: In this example a single aliquot of each of the samples should be aliquot to the defined Assay Plate, whilst the liquid with the tag Standard will be diluted to a Dilution Plate.
