Understanding miniaturized purification resource requirements

To automate a miniaturized purification protocol that uses robocolumns, you need the devices, labware and liquids that follow.

Devices

  • A Tecan Freedom EVO® or Tecan FLUENT® .

  • A Te-Chrom®.

  • A Te-Shuttle.

  • A Te-Chrom waste pump.

  • A plate reader.

Labware

  • Some long or short robocolumns.

Note: Although you must use either long or short robocolumns (not both), each one can contain a different volume of resin

  • A robocolumns array plate.

  • Fixed tips on your liquid handler’s FCA.

  • Some collection plates, to collect fractions.

  • One or more input plates, to contain the samples and buffers at the start of the execution.

Example: A 100 mL trough, a 12-well trough, or a tube.

Liquids

  • The protein samples you want to purify.

  • One or more buffers for each chromatography stage.

Example: A buffer for the equilibration stage, a buffer for the wash stage, a buffer for the elution stage, etc.

Tip: You can use the same buffer for different chromatography stages.
