Tecan Freedom EVO®
Synthace needs to know where the devices and carriers are, which tip types and microplates are on the device, and which liquid classes you have added. To tell Synthace this, your FAS added a Tecan Freedom EVO® configuration to Synthace during its deployment to your organisation. If you need to change your deck layout, add new labware or update your liquid classes for the miniaturized purification protocol, you must add a new Tecan Freedom EVO® configuration to Synthace.
Tip: We recommend that you read this article in conjunction with this one, which explains how to create a new Tecan Freedom EVO® configuration and add it to Synthace.
Worktable
The worktable must contain a Te-Chrom and a Te-Shuttle.
Note: You might set up the deck of your Tecan Freedom EVO® differently, but it still requires this hardware.
If the worktable does not contain a Te-Chrom or a Te-Shuttle, add one from Carriers > Miscellaneous carriers.
If you always want to use plates with deep wells, set your Te-Chrom in a high position, then add a Te-Shuttle transfer/Te-chrom to the worktable.
If you only want to use plates with deep wells some of the time, and might want to use plates with shallow wells at other times, keep your Te-Chrom in a high position, but create a second Tecan Freedom EVO® configuration.
In the second Tecan Freedom EVO® configuration, add a Te-Shuttle transfer/riser to the worktable.
When you want to use plates with deep wells, select the first Tecan Freedom EVO® configuration in Synthace and do not add a riser to the Te-Shuttle.
When you want to use plates with shallow wells, select the second Tecan Freedom EVO® configuration in Synthace and use a riser with the Te-Shuttle.
Robot vectors
During a chromatography stage, if you do not want to collect the liquid that passes through the robocolumns, the Tecan Freedom EVO® will send the liquid to the waste tray instead.
For the device to do this, the ROBOT VECTORS > Robot Vectors section must include a vector that engages the waste tray and a vector that disengages the waste tray.
Note: The name of the vector that engages the waste tray must include the word ENGAGE. The name of the vector that disengages the waste tray must include the word DISENGAGE.
Tips
In the Freedom EVOware® configuration, the tip types must be Fixed.
Click Configure.
On the DEVICES tab, open Instrument options.
Beneath Instrument, select LiHa, then check that the tip types are Fixed.
Labware
The LABWARE > Microplates section must include each microplate that you want to use, and you must link each one to a plate type in Synthace.
Example: We want to use short (200μl) robocolumns, so we add a RoboColumnShort microplate to Freedom EVOware®.
We link the RoboColumnShort microplate in Freedom EVOware® to the RoboColumn Short plate type in Synthace.
Warning: In Freedom EVOware®, the value in the Additional Information field must start with #.
Liquid classes
The LIQUID CLASSES > Custom section must include the miniaturized purification liquid classes.
Note: The miniaturized purification liquid classes have names in the following format.
Flowrate_{x}cm/h_{y}ml/s
(e.g., Flowrate_1000cm/h_55.6ul/s).
Plate reader
Synthace needs to know which absorbance protocol you want the plate reader to use to read the collection plates.
To tell Synthace this, your FAS added a plate reader configuration to Synthace during its deployment to your organisation.
If you want to use a different absorbance protocol for the miniaturized purification protocol, you must add a new plate reader configuration to Synthace.
Tip: If you want the RoMa arm to transfer the collection plates between the deck of the Tecan Freedom EVO® and the plate reader, then automatically run the plate reader tasks, you must add the plate reader as an accessible device. To learn more, click here.
