Configure a Hettich Centrifuge with a Tecan Fluent

Add a Hettich centrifuge as an integrated device on a Tecan Fluent to use in filter plate workflows.

This article walks you through the steps of adding a Hettich centrifuge to the Synthace Base Method in Fluent Control and reconfiguring your Tecan Fluent in SynthaceHub.

- Hettich centrifuge has been installed and taught by a Tecan engineer
- Hettich centrifuge has been added as a device in Fluent Control
- Hettich centrifuge has been added to your worktable in Fluent Control
- Synthace base worktable, script, and method are set in Fluent Control
- SynthaceHub is installed on the Tecan Fluent computer

Warnings

This article does not cover Tecan Fluent teaching for the Hettich centrifuge, labware (including stacked plates), or other devices. Make sure your Tecan Fluent is taught properly before executing your workflow with Synthace. If you need help teaching your Tecan Fluent or installing a Hettich centrifuge, please contact your Tecan representative.

Instructions

1. Add a Hettich Centrifuge to the Synthace Base Worktable

  1. In Fluent Control, ensure the Hettich centrifuge has been added as a device by navigating to the Controlbar menu on the left side of your screen and clicking on Carriers > Device.

  2. Add the Hettich centrifuge to your current worktable in Fluent Control. It should be located underneath the Fluent deck. Make sure it is placed in the correct position for your device.

  3. Create a copy of your Synthace Base Worktable (e.g. "SYNTHACE Base Worktable") by right-clicking on the worktable in the Worktables menu and selecting "Duplicate."

  4. Rename this worktable and click "Save."

  5. Add the Hettich centrifuge to this worktable in the correct location and click "Save."

    1. Be sure to also add any regrip positions to this worktable, or any additional devices, such as a plate reader or shaker.

    2. Do not remove any carriers previously added to this worktable.

  6. Right-click on the Hettich centrifuge and click "Edit" to open the device settings.

  7. Under "General Settings," make sure the "Carrier is device" checkbox is checked. Select the appropriate device (e.g. "Centrifuge") from the "Device" dropdown menu.

  8. If you are adding any additional devices, such as plate shakers, repeat this process by right-clicking each device and selecting "Edit" to open up the device settings.

    1. Make sure "Carrier is device" is checked and the correct device is selected from the "Device" dropdown menu.

2. Configure Hettich centrifuge in Fluent Control

  1. On the Hettich centrifuge, press and hold (~8 sec) the PROG button until the display reads "Machine Menu."

  2. Press the PROG button multiple times until the display reads "->Info," then press the START button.

  3. Press the PROG button a few times until the following parameters are displayed: "Rotor," "NMax," and "R" (centrifuge radius). Record the numbers displayed.

  4. In Fluent Control, open up the Synthace Base Worktable with the Hettich centrifuge included. Right-click the centrifuge to open the device settings.

  5. Under the Hettich device settings, open the "Custom Attributes" tab.

  6. Click on the "+" sign to add new custom attributes. Add the following attributes

    1. SynthaceComponentRole, String, "Rotanta460"

    2. SynthaceRotanta460RpmMax, Integer, "<your recorded NMax value>"

    3. SynthaceRotanta460Radius_mm, Integer, "<your recorded R value>"

  7. Make sure the attribute names match those above exactly before pressing "Save."

3. Set a New Worktable for the Synthace Base Script

  1. In the Controlbar menu, open up the Synthace Base Script by clicking on Scripts > Local > SYNTHACE Base Script. Double-click the script to open.

  2. Click the toggle to the left of "Script Name." Select the new Synthace Base Worktable you made that includes the Hettich centrifuge (and any other added devices).

  3. Click the "Save" button.

  4. Double-check that the base worktable with the Hettich centrifuge included has been added to the base method before continuing.

    1. In the Controlbar, click Methods > Local > SYNTHACE Base Method (or other name). Double-click the method to open.

    2. If the Hettich centrifuge is not displayed on the worktable below, make sure the SYNTHACE Base Script has been added to the method. Next, double-check that the correct worktable was added to the SYNTHACE Base Script and that the script was saved correctly.


Once you have made the require changes to the carriers, worktable scripts and method, you are now ready to generate a device configuration for upload to the device instance in the Synthace platform.
