Tecan Spark Series Plate Reader

Learn how to configure Spark Control to produce data files for use with Synthace.

Introduction

Synthace integrates with Tecan’s Spark Series plate reader by reading the data files generated by the Spark Control software and associating them to a plate in a Synthace execution. Data files must be exported from Spark Control and linked to the associated plates directly.

Data export configuration

In order for Synthace to correctly read the data files generated by Spark Control, some Data handling settings must be configured in Spark Control:

  1. Within Spark Control, set the Destination to: New workbook

  2. Configure the Output → Endpoint as follows:

    1. Export mode: List

    2. List orientation: Horizontal

  3. Configure the Output → Kinetic as follows:

    1. Export mode: List

    2. List orientation: Horizontal

    3. Time: Time per cycle

Supported method strips

Detection modes

The following modes are supported:

  1. Absorbance

    1. Multiple reads per well must be set to Not defined

    2. If pathlength correction is enabled, the Correction factor must be set to Manual

  2. Fluorescence intensity

    1. Multiple reads per well must be set to Not defined

    2. Gain must NOT be set to Extended dynamic range

  3. Luminescence

    1. All configurations supported.

Note that the Scan counterparts to the below detection modes are not currently supported.

Kinetic loops

The kinetic loop strip allows for detection mode readings to be repeated. Kinetic loop strips are supported by Synthace.

Actions

Any action strips included in the Spark Control method will be ignored by Synthace.
