A summary of the automation process

This article summarizes the basic steps that you will follow when you automate a protocol with Synthace.

This guide provides a clear overview of the steps involved in automating a protocol using Synthace, from creating your experimental process to executing it.

Create

The first step is to design your Synthace workflow that can be executed in your lab. There are a number of type of workflow which can be used to create liquid handling instructions in Synthace, the Builder, Cherry Picker, or Form to prepare instructions for your lab devices. The most common method is using the Builder, and so this article will focus on this method of experimental design

Example: The image below shows a builder workflow, consisting of steps to automate a protocol for a simple protein quantification assay using a liquid handler.



Simulate

When you simulate a workflow, Synthace verifies that your lab devices can follow the prepared instructions. It translates these instructions into a series of executable steps, calculating necessary details such as:

  • Input liquids needed

  • Transfer volumes

  • Liquid class selection

  • Labware types needed

  • Well locations for input and output liquids

  • Output liquid concentrations

Simply hit the simulate button to perform this translation.

Preview

Open the simulation in the Simulation Details page and use the Preview tab to visualize the instructions. This helps ensure the instructions will produce the desired outcome.

If adjustments are needed:

  1. Go back to the Builder.

  2. Open and modify the workflow.

  3. Simulate the workflow again.


Start Execution

Once satisfied with the instructions, click the Start Execution button. This creates an Execution entity in Synthace, consisting of executable tasks broken down into stages.



Prepare

Prepare the samples, buffer, and reagents required by your workflow. Use the Reagent Stock Calculator in the Execution page to define master stock solutions, and Synthace will provide preparation instructions.

Check the Simulation Details page for input plate preparation details. Use the Setup tab for volume additions and the Preview tab for deck positions.

Schedule

Transfer your device instructions to your device. The process varies based on your execution mode:

For automated workflows using liquid handlers, click Schedule to send the instructions to the device.

Execute

When ready to run the execution, use SynthaceHub to send the instructions to the applications controlling your lab devices.
